  whole blood assay
Chemiluminescent tests for measuring the real time production of free radicals and other non-radical reactive oxygen species (ROS) produced during the respiratory burst and degranulation of white blood cells (leucocytes).

The cell activation assay, with diluted blood or with isolated cells, measures the activation of the NADPH oxidase system of white blood cells in the diluted blood or in isolated. The NADPH oxidase is the system in some kinds of white blood cells that when activated produces the free radical superoxide which is released outside of the cell.

After the cells are activated they sometimes release enzymes such as myeloperoxidase (MPO). MPO uses some of the superoxide as well hydrogen peroxide derived from the superoxide in other chemiluminescent reactions with Pholasin. The cell activation kit contains all the reagents including substances to activate the white blood cells.

This test is very important in investigations into inflammatory diseases and infection. This assay must be carried out on living cells, either isolated or in a sample of diluted blood. The blood for this test should be no more than two days old.

CONTENTS :

Kit components sufficient for 100 x 200¥ìL tests

A. 1 x bottle Pholasin (50¥ìg)
B. 1 x bottle Adjuvant-K¢â (reconstitute to 5mL)
C. 1 x bottle fMLP (reconstitute to 2.5 mL)
D. 2 x bottles PMA (reconstitute to 2.5 mL)
E. 1 x bottle Reconstitution and Assay Buffer (25mL)
F. 2 x bottles Blood Dilution Buffer (50mL)
G. 2 packs of 25 blood dilution tubes and caps
H. 1 x 96 well white luminometer microplates

The assay has many applications:
  • Identify if cells have been activated.Identify cells that may have been primed.
  • Show enhanced luminescence response to the receptor stimulant fMLP during infection.
  • Preview the way cells, circulating in the peripheral blood, are likely to respond at a site of inflammation.
  • Identify abnormalities in the leucocytes of so-called ¡®normal¡¯ control subjects.
  • Be used in drug assessment and discovery.
  • Measure superoxide to fmol concentrations.
  • Identify patients at greater risk of complications to surgical procedures or exposure to materials in medical devices such as those used in renal dialysis and cardiopulmonary bypass.
  • Monitor the progress of an inflammatory disease or complication and the response of a patient to drug intervention.
