Listeria Detection Kit contains reagents for detection of Listeria monocytogenes using the end-point polymerase chain reaction (PCR). Four sets of primers are included in the kit: one set of primers produces a 360 bp amplicon from the phosphoribosyl pyrophosphate synthetase (prs) gene that is common to all Listeria species (not specific for L. monocytogenes); two other sets of primers produce an outer amplicon of 399 bp and a nested amplicon of 294 bp from the Listeriolysin gene (HLY) that is specific for all strains of Listeria monocytogenes; and the last set of primers produces a 391 bp amplicon from the gene encoding the cell wall teichoic acid glycosylation protein (GTCA gene), which is specific for strain 4b of L. monocytogenes. Strain 4b is responsible for most cases of human illness. The names of the primers are shown in the chart that lists the kit components. Kit Features - Kit contains reagents for 100 experimental PCRs plus 10 additional PCRs that can be used for positive control reactions (40 uL per reaction).
- PCR components are provided as a 5X Ready-to-Load MasterMix.
- Forward + Reverse PCR primers are supplied as mixtures for user convenience.
- Outer and Nested Primers are provided for detection of Listeria monocytogenes.
- Primers are provided for specific detection of Strain 4b of L. monocytogenes.
- Sufficient amounts of each primer pair are provided for up to 100 PCRs, to allow maximum user flexibility in use of the kit.
- Molecular Weight marker is included in the kit for sizing of amplicons from experimental samples. The sizes of the 4 largest mw marker bands are 955 bp, 585 bp, 341 bp, and 258 bp.
Assay results are available in approximately 2.5 - 3 hours, depending on thermocycle profile. #1202-01 : with extraction
